CagriSema (Cagrilintide + Semaglutide) for Fat Loss

Investigational once-weekly fixed combination of the amylin analog cagrilintide and the GLP-1 agonist semaglutide, studied in the Phase 3 REDEFINE program for weight management.

Why CagriSema (Cagrilintide + Semaglutide) for fat loss?

According to PubMed, REDEFINE 1 (a 68-week Phase 3a trial, NCT05567796) reported an estimated mean body-weight change of -20.4% with cagrilintide-semaglutide versus -3.0% with placebo, with gastrointestinal adverse events in about 80% of the combination group, mostly transient (Garvey WT et al., N Engl J Med 2025). The earlier Phase 1b trial characterized the once-weekly pharmacokinetics supporting weekly dosing (Enebo LB et al., Lancet 2021). The combination is investigational and not yet approved.

Fat Loss context: Reducing body fat percentage through appetite regulation, lipolysis, or metabolic upregulation. CagriSema (Cagrilintide + Semaglutide) fits this goal because its category (Metabolic) and benefit profile align with the underlying mechanism.

Benefits relevant to fat loss

  • Substantial weight loss in Phase 3 trials (about 20% at week 68 in REDEFINE 1)
  • Greater weight loss than either component alone in the pivotal trial
