SLU-PP-332

SLU-PP-332 is a synthetic agonist of the estrogen-related receptors (ERR-alpha, beta, gamma), orphan nuclear receptors that regulate mitochondrial biogenesis and oxidative metabolism in skeletal muscle. By activating ERR-alpha it reproduces parts of the transcriptional program normally triggered by aerobic exercise, increasing oxidative muscle fibers and endurance in mice. Its metabolic effects in rodents (reduced fat mass, improved insulin sensitivity) follow from this exercise-mimetic activity. Human relevance is unknown.

Scientific Overview

According to PubMed, SLU-PP-332 is a synthetic pan-ERR agonist (highest potency at ERR-alpha) that induced an acute aerobic-exercise genetic program and increased exercise endurance in mice (Billon C et al., ACS Chem Biol 2023), and reduced obesity and improved insulin sensitivity in mouse models of metabolic syndrome (Billon C et al., J Pharmacol Exp Ther 2024). All evidence is rodent; there are no human data.
